NLRP3 inflammasome in alveolar bone loss

Abstract
The NLRP3 inflammasome acts as a sensor of the intrinsic immune system, and it has also been introduced as a mediator of metabolic and inflammatory bone diseases. In oral tissues, many extrinsic and intrinsic factors activate NLRP3 inflammasome to produce the release of IL-1¥â and IL-18 and pyroptosis cell death. Subsequently, these responses adversely affect the main bone cells that regulate alveolar bone homeostasis and result in alveolar bone loss. Based on these findings, there has been much focus on the development of drugs for controlling inflammatory alveolar bone loss, by inhibiting the activation of the NLRP3 inflammasome. In this review, we will discuss the role of NLRP3 inflammasome in alveolar bone loss and the therapeutic potential of NLRP3 inflammasome inhibition.
